TKIs are medications that treat certain types of cancer by blocking specific proteins that help cancer cells grow and multiply. In chronic myeloid leukemia (CML), a genetic change causes the body to produce an abnormal protein called BCR-ABL1, which causes white blood cells to grow out of control. TKIs work by turning off this protein, helping to stop the spread of leukemia cells. Drugs like imatinib, dasatinib, nilotinib, bosutinib, and ponatinib are commonly used to treat CML.

CML occurs when chromosomes 9 and 22 are rearranged. This rearrangement creates the BCR-ABL gene, which results in the production of a protein that signals uncontrolled white cell growth. There are several risk factors that could increase a person’s risk of developing CML, such as:

High radiation exposure is associated with an increased risk of CML, especially those who have been exposed to nuclear accidents and atomic bomb explosions.

CML is more common in men than women, though the reason why remains unclear.
